Abstract

VEGF is a pivotal stimulatory growth factor in tumor-induced angiogenesis. The first clinical effort in our department to stop tumor progression concerned inhibition of VEGF function. Clinical experience with the inhibitor of VEGF-type 2 receptor phosphorylation, SU5416, started in July 1999 and has been closed recently. All the data are in preparation for publication. Recently, a phase I trial of the tyrosine kinase inhibitor SU6668, given as an oral formulation has been initiated. SU6668 is a tyrosine kinase inhibitor affecting not only VEGF-r function, but also the function of bFGF-r and Platelet-Derived Growth Factor-r (PDGF-r). Since November 2000 a phase I trial of recombinant human endostatin has been started. Endostatin is administered by continuous infusion followed by subcutaneous administration. In both protocols extensive additional data are being collected.
