Novel strategies for controlling and applying homologous recombination in plants, part Genome Editing

The project will provide proof of principle for the applicability of the power of zinc fingers (ZFs) in genome editing in a polyploid, economically important food crop by means of zinc finger nucleases (ZFNs). ZFNs will be used to study whether the allelic composition of a polyploid target genotype can be changed into a desired configuration and whether this works for one or all alleles. The possibility to introduce homozygosity for a specific allele through ZFNs will also be investigated. This part on Genome Editing will be carried out in close collaboration with the sister -project on Genome Interrogation in a model crop by means of ZF-based artifical transcription factors (ZF-ATFs) from IBL, Leiden.
